Summary The purpose of this study was to determine the effects of ACE inhibition on nuclear proliferation and hypertrophy in overloaded skeletal muscle. In a 2 x 2 design, adult female Sprague Dawley rats were placed into one of four groups (n = 8/group): 7- day skeletal muscle overload (O), sham-operation (S), 7-day skeletal muscle overload with ACE inhibition (OA), or sham operation with ACE inhibition (SA). Functional overload of the plantaris and soleus muscles was produced by bilateral surgical ablation of the synergistic gastrocnemius muscles, while ACE inhibition was accomplished via the addition of the ACE inhibitor enalapril maleate (0.3 mg/ml) to the animals' daily drinking water 2 days prior and 7 days post-surgery. Cross-sections of plantaris and soleus muscles from all groups were immunohistochemically stained for dystrophin and bromodeoxyuridine (BrdU). As expected, satellite cell proliferation was significantly higher (p < 0.05) in O compared to S in both the soleus and plantaris muscles. ACE inhibition attenuated this increase in OA soleus but not OA plantaris muscles. Extra-sarcolemmal nuclear proliferation was significantly higher in O compared to S in both the soleus and plantaris muscles, but again only attenuated with ACE inhibition in the OA soleus and not the OA plantaris muscle. Plantaris muscle protein content, wet weight, and fiber cross-sectional area were all increased in O compared to S, and attenuated in OA 7 days post-surgery. There were no differences in soleus muscle total protein content or fiber cross-sectional area after 7 days between any of the groups, but there was an increase in soleus muscle wet weight in the O compared to the S that was attenuated in OA. Collectively, these data indicate that ACE inhibition may attenuate proliferation of satellite cells and other nuclei in overloaded skeletal muscle, but that this effect may depend upon the muscle and/or predominant fiber type being studied.
