Summary Classical MHC class II glycoproteins present peptides to T cells. Nonclassical class II glycoproteins mediate the unloading and loading of peptide from classical MHC class II. Classical MHC genes are usually highly polymorphic. Nonclassical class II genes have little or no variability. In fish, a classical MHC class II Zi-like transcript has been identified, DAB, as well as a divergent MHC class II 5-like transcript, DXB. In the two species of Xiphophorus fishes studied here, X. multilineatus and X. pygmaeus, DAB and DXB were isolated. DAB was highly polymorphic, and the 51 region of DAB, nonsynonymous and synonymous substitutions were found to have around a ten-fold increase versus the remainder of the transcript. DXB had very little variability. Alternative splicing of the DXB transcript was observed, but not of DAB. Two alternative splice patterns were found; a 16 codon deletion and a 5 nucleotide deletion that leads to an extension of the transcript. The alternative splice pattern, lack of polymorphsim, and divergence of DXB from DAB suggest that in fish, DXB may serve in a nonclassical MHC class II role and DAB in a classical roles.
