Subject Hox genes are a regulated set of transcription factor genes that determine cell fate and function as a developmental "master switch" in the determination of cell fate and tissue identity during embryogenesis (Wei-Feng et al., 1989). The Homeobox encodes the homeodomain, a 60 amino acid residue, which has a helix-turn helix motif that superficially resembles the DNA binding domains of prokaryotic repressor proteins. Mammalian Hox genes are clustered in restricted regions of the genome on four distinct chromosomes that presumably evolved by duplication of a primordial gene cluster. It has been recently demonstrated that zebrafish, a teleost fish from the order cypriniformes, possesses 7 Hox clusters (namely aa,ab,ba,bb,ca,cb and da) that contain a total of 48 genes, including one pseudogene. It is hypothesized that another round of duplication occurred in the teleost after the divergence of ray-finned and lobe finned lineages about 420 million years ago. In the case of the zebrafish, that duplication event was followed by the loss of a complete cluster (Amores et al., 1998). Our research has identified and characterized the expression pattern of two Hox genes from the Ca cluster, C5a and C6a, in Morone saxatilis using in situ hybridization. It was hypothesized that the expression pattern of the Hox C5a and C6a in Morone saxatilis would be similar to that observed in zebrafish embryo. This research confirmed homology between C5a expression in zebrafish and striped bass but did not identify any homology between C6a expression in the two species. Hox C5a expression was observed in the neural tube, otic vesicle, dorsal aorta and in the enteric system. Hox gene C6a expression was observed in the otic vesicle region exclusively. Hox gene expression in the otic vesicle region, to this point, has not been described in the literature.
