Summary The various forms of leukemia will be diagnosed in approximately 31,500 people this year in the United States alone. The total lives taken by this disease will exceed 21,500 by the end of this year. Even though new treatments to control this form of cancer are showing promise, there is still much work to be done to elucidate the mechanisms that cause leukemias to occur. Growth and proliferation of peripheral blood cells is strictly controlled by small molecular weight proteins called cytokines. Cytokines exert their effects on blood cells by binding to receptors on the cell surface causing activation of signal transduction cascades. Many forms of leukemia have exhibited overexpression and extended activation of cell signaling molecules. This study examines the ability of Raf kinases, important members of the aforementioned signal transduction cascades, to cause changes in the cell that may result in cytokine independent growth. The ability of conditionally active ARaf isoforms to affect expression of molecules involved in apoptosis, induce autocrine growth, and phosphorylate other signaling molecules was examined. Cross talk between Raf and other important pathways was also observed and assessed. The use of signal transduction pathway inhibitors to reduce cytokine independent growth was examined as a possible pathway specific means of chemotherapeutic intervention.
