| Summary | The 18S and 28S ribosomal genes are usually found in the same chromosome location within a species. However, rDNA genes have been shown to be polymorphic for chromosome location in a number of species, including Hyla chrysoscelis. The mechanism by which these genes 'jump' from one chromosome to another is unknown. Sequencing the ribosomal gene array is the first step towards understanding the nature of this polymorphism. A 3.7 Kb PCR product was generated, cloned and sequenced. BLAST analysis revealed that the product had extensive homology to the 28S rDNA gene of many species, with the greatest homology to Xenopus laevis. Fluorescent in situ hybridization using the cloned PCR product as a probe and pXlr11, a recombinant plasmid derived from Xenopus laevis, as the control probe showed hybridization signal on the same chromosomes and in the same loci with both probes. Approximately 1200 bases have been sequenced; the first 179 bases of the 5' end and 1033 bases near the 3' terminus of the gene. |
