880 3 6| 520-00a| Proteins of the Bcl-2 family are key regulators of apoptosis. Bcl-2, the anti- apoptotic prototypical member of this family, has been localized to mitochondria and is hypothesized to play a role in cell proliferation and may affect the mitochondrial membrane potential, ΔΨ. Many cancer cells express high levels of Bcl-2. In addition, some cancer cells have also been shown to slow their proliferation when induced to overexpress Bcl-2, and some exhibit a high ΔΨ as detected by retention of fluorescent lipophilic cationic dyes. It was therefore hypothesized that the exogenous addition of Bcl-2 to MCF-10A cells, an immortalized human mammary epithelial cell line which expresses little Bcl-2, would result in decreased proliferation and a higher fraction of cells resting in the G0/G1 phase of the cell cycle, and a higher ΔΨ as measured by retention of rhodamine-123 (Rh-123). A stable MCF-10A cell line expressing Bcl-2, called MCF-10A/Bcl-2, was constructed by retroviral infection of the cells with a Bcl-2 construct driven by a murine viral promoter. Cells expressing the construct were selected in the presence of G418, a neomycin analog. Western blot analysis confirmed the presence of Bcl-2 in the MCF-10A/Bcl-2 cell line. Analysis of MCF-10A/Bcl-2 cell proliferation by the MTT proliferation assay revealed a decreased proliferative capacity over parent MCF-10A cells and that of MCF-10A cells infected with an empty vector. Flow cytometric analysis of the cell cycle revealed that a majority of MCF-10A/Bcl-2 cells were in the G0/G1 phase of the cell cycle, with very few in the S phase fraction. Flow cytometric analysis of using Rh-123 indicated that the ΔΨ of MCF-10A/Bcl-2 cells was higher than that of the parent MCF-10A cells and that of MCF-10A cells infected with an empty vector. The results suggest that Bcl-2 may act to inhibit cell proliferation possibly by lengthening the G0/G1 phase of the cell cycle, and that Bcl-2 may be functioning as an ion transporter that regulates ΔΨ of the cells, or as a regulator of the ion transport responsible for ΔΨ.;
