Summary Cux-1 is a member of a family of murine homeobox genes that are structurally related to Drosophila cut. It has been reported that Cux-1 expression is stage dependent in the developing kidney, and that Cux-1 down-regulation may be critical for normal kidney development. To test this hypothesis that Cux-1 down-regulation is necessary for normal kidney development; we have generated and examined transgenic mice constitutively expressing Cux-1 using the CMV immediate early gene promoter. Transgenic mice were generated by pronuclear injection, and progeny that were positive by Southern blot analysis were used to establish permanent lines. Northern blot analysis revealed that three independent transgenic lines express the transgene in the adult kidney. Compared to wild type littermates, the transgenic mice have adult kidneys that are 30-50% larger and also are hyperplastic. In addition, the transgenic mice have tubule-like epithelium lining Bowman's capsule in place of the normal epithelium. Our findings show that persistent expression of Cux-1 results in hypercellularity and abnormal nephron architecture in this mouse mode.
