DNA sequence analysis of two Hox genes in the teleost Morone saxatilis : implications for the evolution of the Hox gene family in vertebrates / by Thomas A. Giesler.

Summary Biologists have long be interested in systems that are responsible for the regulation of development and how they evolved. Of great importance in this regard is the discovery of the vertebrate Hox genes, a group of genes responsible for embryonic development. Hybridization analysis of numerous vertebrate and invertebrate species has revealed the presence of Hox genes in each of the organisms studied, indicating that this class of genes is evolutionarily ancient. In mammals Hox genes are located on separate chromosomes in four distinct clusters, in which the clusters are presumed to have been derived from the duplication of a single ancestral cluster. The purpose of this investigation was to determine if Morone saxatilis (striped bass), a nonmammalian vertebrate, has a similar complement and organization of Hox genes to that of the mammals. We obtained the complete DNA sequence from a bacteriophage lambda-EMBL 3 derived clone prepared from Morone saxatilis genomic DNA. Clone 7H was found to contain two Hox genes, located approximately 7.5kb apart. These two genes were found to be homologous to the known mammalian Hox genes HoxA5 and HoxA7, and were located on the clone in the same order relative to each other as HoxA5 and HoxA7 are in the mammals. Despite the linkage of Morone saxatilis HoxA5 and HoxA7 genes, I was unable to identify a gene corresponding to HoxA6 in the intergenic region between HoxAS and HoxA7. The apparent absence of a HoxA6 gene in the region between HoxA5 and HoxA7 in Morone saxatilis, a representative of the perciform fish, demonstrates that Hox gene evolution in this teleost differs from that in other characterized vertebrates.
