| Summary | The [beta]₂-microglobulin ([beta]₂m) molecule associates with the major histocompatibility complex (MHC) class 1 molecule to present endogenous peptides to CD8' T-cells. In the periphery, these cytotoxic T lymphocytes respond by lysing altered self cells such as those infected by a viais or those undergoing neoplastic growth. In the channel catfish, Ictalurus punctatus, the complete genomic and a full-length cDNA clone of [beta]₂m were isolated and sequenced. Genomic sequences are also reported here for additional ictalurids, including: Ictalurus furcatus, Ictalurus pricei, Ictalurus lupus, Americus natalis, and Americus catus. In the six catfish [beta]₂m gene sequences communicated here, as in zebrafish, two introns separate exons encoding translated amino acids. The [beta]₂m gene of all four Iclalurus species sampled shows a unique N-linked glycosylation signal sequence at the codons for amino acids 90-92 that is absent in both Americus species. Southern hybridization suggests that only one non-polymorphic [beta]₂m locus is present in the genome of I. punctatus. Three hundred eighty-four bp of mitochondrial cytochrome b were also sequenced in these six catfishes. Both the cytochrome c and [beta]₂m sequence data were used to calculate genetic distances between these species. These data yielded a phylogenetic tree consistent with the most recent morphologically-based phylogeny of the Ictaluridae. Sequence data from [beta]₂m intron 1 and the mitochondrial cytochrome c sequence corroborate Ictalurus pricei and I. lupus being more closely related to each other than to I. punctatus. This is the first data suggesting a resolution of the natural history of these three catfishes. |
