| Summary | The Major Histocompatihility Complex (Mhc) class II A genes in the channel catfish (Ictalurus punctatus) were isolated utilizing cDNA libraries and polymerase chain reaction techniques. Two unique clones were isolated from a cDNA library using striped bass and channel catfish Mhc class II A probes. Upon sequence analysis, the two clones were found to have a high degree of homology with the class II A genes from zebrafish and several other teleosts. Primers were designed and two additional clones were isolated by PCR+1. Comparison with other Mhc encoding genes revealed that the products contained the entire coding region. The identification of four different class II A gene sequences in a diploid organism suggests the presence of at least two loci, results that are supported by Southern blot analysis. Southern blot analysis also confirmed a greater degree of polymorphism in the B genes as opposed to the A genes. When the four gene sequences were translated, each contained several structural motifs that would indicate function, including the conserved cysteines within the a1 and a2 domains. |
