| Summary | The present study partially characterizes the terminal carbohydrate moieties of glycoconjugates during limb regeneration using lectin histochemistry, and examines the distribution of "neural elements" using Bodian histochemistry in regenerating limbs treated with Triap and/or retinoic acid (RA) at day 46 post amputation. A panel of eleven horseradish peroxidase conjugate (HRP) lectins were used to probe epidermal glycoconjugates for specific carbohydrate moieties. The eleven lectins utilized are LTA, UEA-I, DBA, GSAI-B4, GSA-II, Con A, Scon A, WGA, PN°, and ECA. Most regenerating tissues exhibited affinity for seven specific lectins by retaining their binding characteristics at the end of the 46-day experimental period. Tissues treated with Triap and/or RA revealed similar terminal carbohydrate moieties of glycoconjugates and the distribution of "neural elements". Extracellular matrix of the regenerating limbs contributed in directing regeneration. These findings demonstrate that Triap and/or RA brought about integumental changes during limb regeneration in the red-spotted newt. |
