520 3 a| Prolactin (PRL) dependent secondary sexual characteristics in the male red-spotted newt include an expanded tail fin and the presence of nuptial pads on the medial aspect of the hind limbs. During unfavorable environmental conditions such as high temperatures and conditions of low water and/or hypoxia, the secondary sexual characteristics of the red-spotted newt regress. Dopamine (DA), a catecholamine is secreted in times of "stress" and is a potent inhibitor of PRL in many animals, but the role of DA in urodele amphibians is unknown. In the present study, specific DA antagonists (pimozide, fluphenazine, haloperidol, clozaril) are used to selectively block receptors on the lactotrophs of the adeno-hypophysis. Results show that animals treated with DA antagonists retain PRL dependent characteristics longer in colder temperatures. In addition, pimozide treated animals retain tail height and nuptial pads indicating that DA was the PRL inhibitor in this species and was acting via D2, receptors in the anterior pituitary. This study also reveals that pimozide elicited melanocyte-stimulating hormone release in the newts. These results suggest that environmental stress in this species of urodele is mediated, in part, by dopaminergic pathways.;
