Summary The present study examines the integumental morphology and the effects of prolactin in the slimy salamander, Plethodon chlorohryonis, utilizing the techniques of light microscopy and lectin histochemistry. The expression and distribution of glycoconjugates in epidermis and integumental glands were visualized using a panel of nine horseradish peroxidase labelled lectins. The epidermis consists of 3-4 cell layers and exhibits a single layer of stratum corneum which is relatively unkeratinized. Mucous glands, found predominantly on the ventral surface, contain a sulfated, acidic muco-polysaccharide which contained residues of mannose, glucose, and D-GlcNAc. Two morphologically distinct types of granular glands were also identified. The smaller Type 1 glands were most abundant on the dorsum of the trunk, whereas the large Type 2 glands were found exclusively on the dorsal aspect of the tail. Within the Type 2 glands, granules exhibited a gradient in staining with DBA (D-GalNAc) and LTA (fucose). Administration of 3 lU of ovine prolactin on alternate days over a two week period failed to alter integumental lectin binding patterns but appeared to elicit a water drive during the first week of the experimental period.
