Summary This study reports the initial isolation and characterization of Major histocompatibility complex {Mhc) class II (beta) genes in the teleost, Morone saxatilis. Polymerase chain reaction (PCR) and dideoxy sequencing techniques were employed to amplify and characterize seven subclones (Mosa-C-1, Mosa-C-2, Mosa-C22, Mosa-R41, Mosa-S-1, Mosa-S-2, and Mosa-S-3) encoding the entire mature Mhc class II (beta) chain. Striped bass from three different locations (Roanoke River, Chesapeake Bay, and Santee-Cooper Reservoir) were used in this study. Extensive polymorphism was found in the (beta)1 domain encoding region of the cDNA, which corresponds with the functional peptide binding region (PBR) of known Mhc class II molecules (Figure 4). Variable amino acid residues in the (beta)1 domains from the striped bass clones correspond with polymorphic sites of (beta)1 domains from other teleosts and higher vertebrates. Other regions of the striped bass Mhc class II translated proteins correlated well with those of known vertebrate Mhc class II proteins. Seventy-one percent of the variability within the presumed PBR of the striped bass Mhc class II (beta) chain gene was found to correspond with that of the PBR of a human Mhc class II (beta) chain gene (Brown et al. 1993).
