Summary This thesis reports the isolation and characterization of a cultivable anaerobic, cellulolytic microflora from the gastrointestinal (GI) tract of the pinfish, Lagodon rhomboides. Anaerobic sampling and culturing techniques were employed in isolating forty carboxymethylcellulose (CMC) digesting obligate anaerobes from the GI tract of L. rhomboides. CMC is a water soluble derivative of cellulose. Based on phase contrast microscopy. Gram stain reaction, biochemical tests, and gas chromatographic analysis of volatile fatty acid (VFA) and organic acid production, the obligately anaerobic carboxymethylcellulase (CMCase) producing bacteria isolated from the GI tract of L. rhomboides have been assigned to three distinct taxonomic groups, designated I, II, and III, representing 92.5% of the CMCase isolates. Group I isolates are non-motile spore forming bacilli, presumptively placed in the genus Clostridium. Isolates assigned to group II are motile, spore-forming bacilli also presumptively placed in the genus Clostridium. Group III isolates are non-motile, non-spore forming coccobacilli that can not be assigned to any known genera. Based on this study, all of these obligately anaerobic, cellulolytic bacterial isolates belong to unknown, hitherto undescribed new species.
