| Summary | The purpose of this research was to evaluate the in vivo effect of a prolonged hyperadrenalinaemic state in rats on GLUT-4 glucose transporter protein and the messenger ribonucleic acid (itiRNA) expression. Epinephrine is an important hormone in the control of glucose metabolism and homeostasis of blood glucose. The effects of prolonged epinephrine exposure were examined by Budohoski and co-workers (1987), and they demonstrated improved glucose tolerance and insulin sensitivity after 72 hours of stimulation. In light of this observation, and due to the lack of information concerning chronic epinephrine exposure on GLUT-4 expression in skeletal muscle, the hypothesis was proposed that chronic stimulation with epinephrine would alter GLUT-4 expression in skeletal muscle. Rats were implanted with epinephrine releasing tablets for one and five days. Upon euthanasia, trunk blood was collected for plasma assays of epinephrine, glucose, and insulin. Muscle samples were examined for GLUT-4 protein content by Western blot analysis and GLUT-4 mRNA was calculated along with ratios for GLUT-4 and B-actin mRNA by Northern blot analysis. Plasma epinephrine and glucose levels were significantly higher in both experimental (epinephrine implant) groups. The GLUT-4 protein content was reduced by 37-40% (P[less-than]0.05) in both red and white muscle fiber-types in the one day epinephrine treated animals. The five day exposure showed no change in GLUT-4 protein. GLUT-4 mRNA showed a similar trend, with the 1-day exposure in the red muscle fiber-type being reduced by 42% (P[less-than]0.01) However, interpretation of the mRNA data was clouded by a decrease (48%)in the constituitive message for B-actin at one day of epinephrine treatment. By five days of exposure, the mRNA content was that of the control groups for both fibertypes. |
