Summary Recent literature suggests that Cyclosporin (CsA) administration results in an immediate, significant increase in serum prolactin (Cardon et. al., 1984). The purpose of this study was to determine if the reported results of CsA treatment are a result of direct action at the prolactin receptor level. Serum prolactin (Prl) and luteinizing hormone (LH) concentrations were used to determine the effect of CsA treatment in vivo in adult Sprague-Dawley rats. Acute CsA treatment following a dose response schedule (0.12, 1.2, 12.0 or 120.0ug/100g bw) resulted in a significant increase in serum luteinizing hormone at a CsA dose of 1.2ug/100g bw i.p. one hour after treatment as compared to controls. There was no change in trunk blood serum prolactin. Repeated CsA treatment at one of four graded doses i.p. resulted in increases in serum LH which were independent of dose. The same treatment had no effect on trunk blood serum Prl in intact male rats or ovariectomized female rats after a 24 hour treatment period. Oral CsA treatment (12.0ug/100g bw) daily for 21 days in male rats had no effect on serum LH when measured on days 7, 14, and 21, nor did it affect pituitary LH content on day 21. Serum Prl and pituitary Prl were also unchanged on day 21. Serum creatinine levels were significantly lower in the CsA group as compared to untreated controls. Chronic high dose oral CsA treatment (1.5mg/100g bw) daily for 21 days in male rats had no effect on serum or pituitary gland Prl concentration. Serum LH increased significantly at day 14 but was within normal limits by day 21. Pituitary LH content increased significantly in response to CsA treatment. In addition, serum creatinines were significantly elevated in the experimental group. In summary, CsA has a stimulatory effect on LH secretion by the anterior pituitary which is independent of dose. This effect may be mediated by both direct action at the gonadal level and indirect action at the hypothalamic level. CsA has no effect on serum Prl or pituitary Prl content when examined under the wide range of experimental designs used in this project.
