Summary The genetic heterogeneity of mitochondrial DNA (mtDNA) from Roanoke River striped bass (Morons saxatilis) was examined using restriction endonuclease analysis. Mitochondrial DNA isolated from a total of 32 striped bass captured at different times during the 1989 and 1990 spawning runs was digested with a battery of twelve restriction endonucleases. Restriction endonuclease fragment patterns resulting from digestion of mtDNA were analyzed by agarose and polyacrylamide gel electrophoresis. Five of the restriction endonucleases examined (Acc I, Nde I, Spe I,Dde I, and Taq I) revealed the existence of restriction fragment site polymorphisms among the 32 fish. Restriction fragment site polymorphisms resulting from Acc I and Nde I digestion both occurred in the same group of eight (8) striped bass. Restriction fragment site polymorphisms identified by digestion with Taq I, Spe I, and Dde I all occurred in separate groups of striped bass. Restriction endonuclease analysis also revealed the presence of five distinct restriction fragment size polymorphisms among the 32 fish. We were unable to demonstrate a correlation between the appearance of restriction fragment site polymorphisms and restriction fragment size polymorphisms in the striped bass examined in our study, suggesting a need to reevaluate results that rely solely on restriction fragment size polymorphisms as a stock identification method. Based on the discovery of five restriction fragment site polymorphic classes of striped bass, we conclude that considerable genetic heterogeneity exists in the Roanoke River and that mtDNA analysis may prove extremely useful in striped bass stock identification efforts.
