Summary Invertible DNA segments are known to exist in several microbial genomes. Inversion occurs by site specific recombination within the inverted repeat sequences flanking these segments. Efficient site specific inversion results from joint activities of the recombinase, other as yet poorly characterized proteins, and enhancer sequences. The recombinases and recombination sites of bacteriophage PI, bacteriophage Mu, Salmonella, and E. coli are highly homologous and mutual complementation has been shown. The plasmid, pKZ601, (derived from bacteriophage PI) has an invertible segment with the recombinase gene between cix sites flanked by abbreviated inverted repeats. Normal populations of pKZ601 DNA contain equimolar concentrations of both invertible segment configurations shown by electrophoresis of restriction fragments resulting in four equimolar bands. Two spontaneous mutations, one exhibiting no inversion, and another with non-equimolar inversion have occurred in pKZ601. Studies performed on the non-inverting mutant (pKZ60S) did not conclusively characterize this mutation. DNA thought to contain a second non-inverting mutant, pKZ603, was checked by restriction fragment analysis of 36 transformants and a non-inverting mutation was not detected. Some transformants, however ¡, shewed a non-- equimolar fragment distribution. Subsequent serial transformations showed variation between equimolar and non-equimolar distribution phenotypes. A hypothesis that this phenotype is due to a mutation in the recombinase promoter was tested and eliminated.
