| Summary | A type of site-specific recombination known as inversion can control gene expression by determining which gene or set of genes is transcribed. Inversion of the C segment of bacteriophage P1 varies the expression of tail fiber proteins, thus determining the host range. Inversion of the 4.2 Kb segment is mediated by a recombinase encoded by the cin gene located directly adjacent to the C segment. The cin promoter has been localized within a 100 bp region upstream of the coding sequence of the gene. This region, in addition to the gene, has previously been sequenced, revealing several sequences bearing some homology to consensus promoter sequences. Using a plasmid which contains a functional inversion system, this study involved creating mutants by forming deletions into the promoter region of cin. One mutant was analyzed in which the deletion extended past the start codon, completely abolishing inversion. Another mutant was isolated in which the deletion extended past the ribosomal binding site. This mutant exhibited a low, but detectable, rate of inversion due to transcription of cin from an undetermined promoter. Three mutants were isolated which exhibited a somewhat lower rate of inversion than that of the parent plasmid. Analysis of these revealed that the -35 region of three possible promoters had been deleted in each, raising the possibility that the -35 sequence of the cin promoter is not essential for transcription. Alternate hypotheses are addressed. |
