Summary The purpose of this study was to determine if free fatty acids cause detachment of lipoprotein lipase from the endothelial bed of blood vessels in vivo. Free fatty acid levels were raised in vivo by three different ways: Induction of diabetes by streptozotocin injection, exhaustive exercise, and injection of oleic acid through tail vein. In diabetic rats, plasma glucose, triglycerides, and free fatty acids were highly elevated. Exercised rats show a significant decrease in serum glucose and triglyceride levels, but free fatty acid levels were not different from controls. Lipoprotein lipase activity was markedly reduced in the diabetic-fed and exercised rats compared to controls. Adi pose tissue lipoprotein lipase activity was markedly reduced in the diabetic and exercised rats compared to the respective controls. No significant change in either muscle or heart lipoprotein lipase activity was observed. Furthermore, oleic acid injections could not raise the serum free acid levels as expected; however, serum triglyceride levels were elevated with higher doses of oleic acid. From in vivo studies, no correlation between serum free fatty acid and lipoprotein lipase activity could be established. This suggests that free fatty acids do not detach lipoprotein lipase from the endothelial bed of vessels. A more definitive approach to study the effects of free fatty acids on the dislodging of lipoprotein lipase from the endothelial bed was used. Perfusing rat hemicorpus preparations containing only epididymal fat pads and hind muscles with oleic acid albumin complex (7:1 molar ratio) failed to show higher lipoprotein lipase activity in the perfusate fractions, whereas heparin perfused hemicorpuses showed a significantly higher lipoprotein lipase activity in the perfusate. No change in lipoprotein lipase activity of adipose or muscle tissues of hemicorpuses perfused with either oleic acid or heparin resulted. Based on the in vivo and perfusion studies, these results strongly suggest that the changes in tissue lipoprotein lipase activity are-independent of elevations in the free fatty acid levels and that elevated free fatty acid levels may not have any significant effects on the decrease in lipoprotein lipase activity in the tissues of diabetics.
