Summary Versican is a hyalectin proteoglycan highly expressed in the extracellular matrix of precartilage mesenchymal cells and presumptive joints of the embryonic limb. In previous experiments, versican expression has been linked to condensation of precartilage mesenchymal cells in vitro, however, versican's role within the limb skeletal template has yet to be tested in vivo. Thus, we hypothesized that expression of versican within the embryonic chick limb in ovo would facilitate chondrogenesis. Our lab assessed the effects of over-expression of the versican carboxy terminal domain, G3 during chick limb development in ovo through injection of G3 adenoviral constructs generated in our lab. Whole mount alican blue and immunohistochemical staining revealed that over-expression of the G3 domain dramatically increased the size of cartilage elements in the developing embryonic limb and joint region. These results provide valuable insight into versican's role in limb development, as well as the possible independent function for the G3 domain.
