| Subject | The expression patterns of developmentally regulated genes are thought to be controlled by complex networks of cis-regulatory elements. The use of transgenic host vector reporter systems has become a prevalent means of functionally studying these regulatory networks. Analyses of gene regulatory modules using transgenic host-vector reporter systems in animal models such as mice and chicken have provided insight into the complexity of regulated gene expression, but investigations using these models can be costly, labor intensive, and cumbersome. The zebrafish (Danio rerio) has proven to be an excellent model for vertebrate developmental studies, but the lack of efficient host vector systems has made functional transgenic studies very labor intensive, and time consuming, because existing methods require establishment of stable lines, or construction of expression patterns from a composite of mosaic F₀ injected embryos. The Tol2 transposon system is a recent advancement in transgenic technology that has provided a solution to the problem of highly mosaic injected zebrafish embryos. In this study we report the development of a Tol2 transposon-based vector system that provides a level of genome integration and promoter function suitable for conducting enhancer element mapping studies in zebrafish in the F₀ generation, which obviates the need for time consuming and labor intensive construction of stable transgenic lines and affords the opportunity to accelerate the rate of in vivo enhancer mapping in zebrafish. |
