Summary The extracellular matrix (ECM) secreted by mesenchymal cells during endochondral ossification is critical in the specification and differentiation of those cells. Versican is a chondroitin sulfate proteoglycan and previous work in this lab has shown that it is highly expressed in the ECM of precartilage mesenchymal cells and in the presumptive synovial joint. Furthermore, this lab has shown that morpholino-mediated knock-down of endogenous versican expression during chick limb development compromises chondrogenesis. The mechanism of versican's function during limb development is yet to be elucidated. While much of versican's effect has been attributed to its glycosaminoglycan chains, the individual domains of versican have been shown to possess distinct and sometimes opposing functions when expressed ectopically. The current study sought to determine if over-expression of the G1 domain and V3 isoform of versican in ovo would result in independent and diverse effects on chick limb development, possibly implicating discrete areas of versican in different aspects of limb skeletogenesis. An adenovirus encoding these molecules was microinjected into the prechondrogenic chick limb bud which resulted in a thickening of the skeletal anlagen implicating both of these molecules in early stages of limb development.
