Summary Male Dub:ICR mice were treated with 5-thio-D-glucose at a rate of 35mg/kg/day for either 7, 14, 21, 28, or 42 days. At the end of each treatment period, radiorespirametric studies with glucose-1-14C and glucose-6-14C were done on testis and liver tissue from treated and control mice. C-1/C-6 ratio was calculated to determine the relative activity of pentose phosphate pathway to glycolysis-tricarboxylic acid pathway. The liver tissue showed no significant difference in 14CO2 percent recovery with one exception. A lower recovery from glucose-6-14C during the 14-day period was observed in the treated. There was no difference in C-1/C-6 ratio for the liver. By 42 days of treatment, the percent recovery from glucose-114C was significantly higher in the treated testes. The C-1/C-6 ratio in the same period was also significantly higher, indicating increased pentose phosphate activity in the treated. There was no observable effect of the treatment on the 14CO2 activity derived from glucose-6-14C in the testis tissue.
