| Summary | Prolactin (PRL), growth hormone (GH) and somatolactin (SL) have important functions including postnatal growth, lactation, and osmoregulation. However, the roles of PRL/GH/SL in development remain unclear. The current study examined the involvement of these hormones in zebrafish development. The transcripts of PRL/GH/SL(alpha) were found to be present in the ovary and at the examined stages of development, from one-cell stage to 14 days post fertilization (dpf), although SL(beta) mRNA was absent between one-cell stage and 6 hours post fertilization (hpf). The transcripts of GH, PRL and SLs increased significantly at the time of pituitary gland induction (12-18 hpf) and during larval development, (2 to 5 dpf), suggesting that PRL/GH/SLs might have roles in formation of the pituitary gland and/or embryonic development. The potential actions of PRL/GH/SLs in development were further examined by morpholino antisense knockdown. No phenotypic difference was observed in GH or SL(alpha) knockdown embryos compared to embryos injected with a control morpholino oligonucleotide. In contrast, significant effect on the development of gas bladder was observed in SL(beta) knockdown embryos. Knockdown of PRL not only affected the gas bladder, but also significantly reduced pigmentation, body length and eye size of the embryos. The defects caused by PRL antisense could be rescued with the modified PRL mRNA. The observations suggest that PRL have important functions on zebrafish development. |
