Summary Development of the vertebrate inner ear from a simple epithelial placode to an intricate three-dimensional structure housing the organs of audition and equilibrium is a complex process requiring massive cell movements and differentiations. Normal development of these organs has been associated with extracellular matrix molecules which have yet to be identified. Glycosaminoglycan epitopes found on versican, an extracellular matrix proteoglycan, are distributed in the developing inner ear. Known to participate in various tissue modeling processes, versican is implicated in neural crest cell migration and versican expression has been identified in the brain structures of the auditory pathway. The heart defect {hdf) mouse features a lacZ reporter insertion that disrupts the versican Gene. Hdf transgene expression in heterozygous inner ears was evaluated by lacZ histochemistry, and versican expression on the protein level was evaluated in heterozygous and wild type inner ears of animals from E18/19-P6months. Versican was transiently localized in the condensing mesenchyme and cartilage of the otic capsule, surrounding the axons of cochlear and vestibular nerve, in the efferent and afferent fibers of the receptor organs, in the stria vascularis and in the arcuate spiral limbus. Faint transient expression was seen in sensory epithelium. Versican may participate in morphogenesis, organogenesis and neurogenesis in the developing inner ear by influencing cell-cell and cell-matrix interactions that regulate processes such as cell migration, adhesion, proliferation and differentiation.
