Summary The purpose of this study was to determine how the degree of cardioprotection conferred to two inbred rat strains during ischemia varied depending on the artificially selected phenotype for endurance running. Two cohorts of rats corresponding to the LCR (low-capacity for running) and HCR (high-capacity for running) were randomly divided into two groups each, an ischemia and non-ischemia group. An ex vivo ischemic insult was administered in an incubation tray containing anoxic media for 2 hours. Lactate dehydrogenase (LDH) and creatine kinase (CK) were chosen as biomarkers of ischemia to estimate degree of cardioprotection and clinical assays of these enzymes were performed for both myocardial tissue homogenate and the anoxic, incubation media. Significant findings (P[less-than]0.05)) were reported for LDH and CK enzymes in the incubation fluid; both enzymes were significantly elevated in the LCR phenotype when compared to the HCR phenotype. Two multiplex array panels were utilized to measure pro- and antiinflammatory cytokine profiles. The Lincoplex® rat cytokine/chemokine array was used to quantify cytokine concentrations in both tissue homogenate and the incubation media. Five pro-inflammatory cytokines (IL-la, IL-ip, IL-2, lL-4, and IL-18) reported significant elevations in concentration in LCR versus HCR rats. The results of this study are the first to demonstrate direct evidence of cardioprotection in HCR rats that had only been postulated by previous reports.
