Summary Human immunodeficiency virus type 1 (HIV-1) interacts with two main receptors via HIV-1 envelope (Env) for entry into a target cell. These include the primary receptor human CD4 (hCD4), and a secondary coreceptor; usually either the hCXCR4 or hCCR5 chemokine receptor. During HIV-1 infection there is a gradual depletion of CD4⁺ T cells consistent even during periods of viral latency. Among other theories, the induction of apoptosis, or programmed cell death, is hypothesized as being a means of this subsequent cell death. However, there is still ambiguity as to the mechanism of HIV-1 / receptor interaction and its possible role in the induction of apoptosis. This study, analyzed the interaction of HIV-1 envelope (gp120) with specific chemokine receptors. The chemokine receptors hCXCR4, hCCR5, and hCCR3 were analyzed for their importance in initiating signal transduction and apoptosis, via stimulation with different isolates of HIV-1 gp120. Both human embryonic kidney (HEK) 293T cells and human T cell lines were stimulated with the gp120 subunit of Env alone or with soluble CD4 (sCD4). The analysis of these receptors resulted in the induction of apoptosis primarily in cell lines expressing the chemokine receptor hCXCR4 in the presence of hCD4. Cells expressing hCCR5 and hCCR3 were more resistant to gp120 induced apoptosis. Signal transduction events in T cells, such as influx of calcium and phosphorylation of Erk also depended on the presence of both hCD4 and hCXCR4.
